Genetic characterization and phylogenetic analysis of foot and mouth disease virus vaccine strains and recent field isolate

Foot and Mouth Disease (FMD) control is a national regional responsibility. FMD outbreaks were detected in three farms Port-Said Governorate 2020, although animals these vaccinated with the local polyvalent inactivated vaccine. Samples from tongue epithelium vesicular fluid collected farms, where FMDV isolation was performed on BHK-21 cell culture. Viral RNA extracted virus isolates, vaccine strains then screened for using conventional Reverse transcriptase polymerase chain reaction. The substitution rates phylogenetic relationship between field isolate determined DNA sequence bioinformatics analyses. results illustrate that analysis 1D region of serotype A‑Africa topotype, Genotype IV, closely related to isolated topotype IV by Animal Health Research Institute, 2020 nucleotide similarity ranging 97.1% 99.74%, revealing genetic variation 5.63-7.88% previously Egyptian 2016 2018. High variations locally used strain A Asia lineage Iran‑05, major antigenic sites VP1 difference 26.34%. Depending findings, Veterinary Serum Vaccine Institute produced an emergency monovalent newly strain, we recommended adding it subsequent prepared batches.
